چکیده

A novel compound, 4-amino-4-cyanobutyric acid, was isolated and identitled from cultures of an unidentified psychrophilic basidiomycete. Triple labeling experiments in vivo involving the simultaneous administration of KW15N and succinic semialdehydeJ4C to fungus cultures showed that these compounds along with ammonia are precursors of 4amino-4-cyanobutyric acid. In addition, cell-free extracts were able to catalyze this reaction. That 4-amino-4-cyanobutyric acid is a precursor of glutamate was also established by triple labeling experiments as well as with crude enzyme preparations. In the latter case nitrilase activity was demonstrated. Enzymes involved in glutamate and succinic semialdehyde metabolism were found, and on this basis a cycle involving cyanide detoxification is proposed.

The Biosynthesis of Folic Acid I. SUBSTRATE AND COFACTOR REQUIREMENTS FOR ENZYMATIC SYNTHESIS BY CELL-FREE EXTRACTS OF ESCHERICHIA COLI*

The formation of folic acid compounds from pteridines and p-aminobenzoic acid by whole bacterial cells has been studied by Korte et al. (1, 2), who reported that C%xanthopterin (2-amino4,6-dihydroxypteridine) could be incorporated into folic acid by several bacterial species, and by Weygand et al. (3), who found that certain strains of bacteria can be adapted to utilize 2.amino-4-hydroxy-6-form...

Flavoproteins: Structures and Mechanisms Glutamate synthase: a complex iron-sulphur flavoprotein

Glutamate synthase (GltS, EC 1.4.1.13) forms with glutamine synthetase (GS, EC 6.3.1.2, reaction 1) the main route for glutamate synthesis in micro-organisms by catalysing the NAD(P)Hdependent transfer of the amide group of glutamine to the C(2) carbon of 2-oxoglutarate (2-OG, reaction 2). The overall reaction catalysed by GS and GltS (reaction 3) is similar to the reaction of glutamate dehydro...

Natural products as safeguards against monosodium glutamate-induced toxicity

Monosodium glutamate is a sodium salt of a nonessential amino acid, L-glutamic acid, which is widely used in food industry. Glutamate plays an important role in principal brain functions including formation and stabilization of synapses, memory, cognition, learning, as well as cellular metabolism. However, ingestion of foodstuffs rich in monosodium glutamate can result in the outbreak of severa...

Two pathways for glutamate biosynthesis in the syntrophic bacterium Syntrophus aciditrophicus.

The anaerobic metabolism of crotonate, benzoate, and cyclohexane carboxylate by Syntrophus aciditrophicus grown syntrophically with Methanospirillum hungatei provides a model to study syntrophic cooperation. Recent studies revealed that S. aciditrophicus contains Re-citrate synthase but lacks the common Si-citrate synthase. To establish whether the Re-citrate synthase is involved in glutamate s...

Propofol differentially inhibits the release of glutamate, γ-aminobutyric acid and glycine in the spinal dorsal horn of rats

Objective(s): Propofol (2, 6-diisopropylphenol) is an intravenous anesthetic that is commonly used for the general anesthesia. It is well known that the spinal cord is one of the working targets of general anesthesia including propofol. However, there is a lack of investigation of the effects of propofol on spinal dorsal horn which is important for the sensory transmission of nociceptive signal...
